     实(shi)时(shi)(shi)调(diao)控(kong)(kong)细(xi)(xi)(xi)胞生(sheng)命活动(dong)对(dui)研(yan)究(jiu)细(xi)(xi)(xi)胞的(de)(de)生(sheng)理功能(neng)具(ju)(ju)有(you)重(zhong)要价值。由于(yu)光(guang)(guang)(guang)(guang)优异(yi)的(de)(de)时(shi)(shi)空分(fen)辨率(lv),生(sheng)物相容(rong)的(de)(de)光(guang)(guang)(guang)(guang)引发(fa)化(hua)(hua)学(xue)工具(ju)(ju)可用(yong)(yong)(yong)于(yu)原(yuan)位实(shi)时(shi)(shi)调(diao)控(kong)(kong)动(dong)态的(de)(de)生(sheng)命过(guo)程(cheng)。传统的(de)(de)光(guang)(guang)(guang)(guang)去笼方法通过(guo)直接(jie)光(guang)(guang)(guang)(guang)照射(she)底物分(fen)子切断化(hua)(hua)学(xue)键从而释放生(sheng)物活性(xing)(xing)分(fen)子,光(guang)(guang)(guang)(guang)去笼方法近年(nian)来的(de)(de)新(xin)发(fa)展希望实(shi)现如(ru)下重(zhong)要新(xin)特质:一(yi)、通过(guo)廉价易得的(de)(de)可见(jian)光(guang)(guang)(guang)(guang)光(guang)(guang)(guang)(guang)源实(shi)现更好的(de)(de)生(sheng)物穿透性(xing)(xing);二、底物具(ju)(ju)有(you)光(guang)(guang)(guang)(guang)稳定性(xing)(xing)从而无需当(dang)场(chang)制备;三、可定位的(de)(de)特异(yi)性(xing)(xing)光(guang)(guang)(guang)(guang)去笼。相比于(yu)直接(jie)光(guang)(guang)(guang)(guang)照射(she)方法,光(guang)(guang)(guang)(guang)催化(hua)(hua)氧化(hua)(hua)还(hai)原(yuan)反应(ying)使(shi)用(yong)(yong)(yong)能(neng)量较低的(de)(de)可见(jian)光(guang)(guang)(guang)(guang),底物在(zai)不存在(zai)光(guang)(guang)(guang)(guang)催化(hua)(hua)剂时(shi)(shi)对(dui)光(guang)(guang)(guang)(guang)惰性(xing)(xing)因而便于(yu)贮存。但目前(qian)将光(guang)(guang)(guang)(guang)催化(hua)(hua)氧化(hua)(hua)还(hai)原(yuan)反应(ying)应(ying)用(yong)(yong)(yong)于(yu)细(xi)(xi)(xi)胞存在(zai)以下困难:一(yi)、常用(yong)(yong)(yong)的(de)(de)过(guo)渡(du)金属(shu)光(guang)(guang)(guang)(guang)催化(hua)(hua)剂具(ju)(ju)有(you)潜在(zai)的(de)(de)细(xi)(xi)(xi)胞代谢(xie)毒性(xing)(xing),会使(shi)其应(ying)用(yong)(yong)(yong)受到限制;二、细(xi)(xi)(xi)胞内广泛存在(zai)的(de)(de)氧气及各种(zhong)抗氧化(hua)(hua)剂会影(ying)响自由基活性(xing)(xing)物种(zhong)寿命及光(guang)(guang)(guang)(guang)催化(hua)(hua)氧化(hua)(hua)还(hai)原(yuan)反应(ying)效(xiao)率(lv)。

      中国科(ke)学院上(shang)海(hai)有机化(hua)学研(yan)究所生命(ming)有机化(hua)学国家重点实(shi)验室陈以昀课(ke)题组致(zhi)力于(yu)发展新(xin)的生物(wu)相容光化(hua)学方(fang)法(fa)用于(yu)化(hua)学及(ji)生物(wu)学的研(yan)究,主要研(yan)究内容包括新(xin)型光反应(ying)发现及(ji)新(xin)型光化(hua)学生物(wu)学工(gong)具的发展。课(ke)题组前期研(yan)究工(gong)作(zuo)中建立了可见(jian)光引(yin)发氧化(hua)及(ji)还原反应(ying)的生物(wu)大(da)分子相容性(J. Am. Chem. Soc. 2014, 136, 2280Chem. Comm. 2015, 51, 5275),并(bing)发(fa)展了水(shui)相条件下生物活性小(xiao)分(fen)子(zi)的可(ke)见光(guang)释放方法(Angew. Chem. Int. Ed. 2015, 54, 1881Angew. Chem. Int. Ed. 2015, 54, 7872)。近日与(yu)上海交通大(da)学(xue)医学(xue)院徐(xu)天乐(le)课(ke)题组合作,首(shou)次(ci)报道了(le)将常用于荧光(guang)(guang)成(cheng)像(xiang)的(de)有机荧光(guang)(guang)分(fen)子荧光(guang)(guang)素或罗丹明衍生物作为生物相容的(de)光(guang)(guang)催(cui)化(hua)剂,在细胞内利用氧气及抗(kang)氧化(hua)剂通过(guo)光(guang)(guang)催(cui)化(hua)氧化(hua)还原反应(ying)产生过(guo)氧化(hua)氢,进而进行脱硼羟(qiang)基化(hua)反应(ying)去笼释放生物活性分(fen)子(Angew. Chem. Int. Ed. 2018, DOI: 10.1002/anie.201811261),文章第(di)一作者为上海有机所博士研究(jiu)生(sheng)王浩岩(yan)。该(gai)反应使用(yong)家(jia)用(yong)CFL或低(di)能(neng)量蓝(lan)色(se)LED灯作为光(guang)源,在中(zhong)性水溶液(ye)中(zhong)可(ke)以对含有(you)酚、羟基、氨基的生物(wu)活性分子(zi)实(shi)现高效的光(guang)去(qu)笼(long)释(shi)放。该方(fang)法可(ke)以通过(guo)光(guang)释(shi)放异丙基-β-D-硫代(dai)半乳(ru)糖苷(gan)分子(zi)来光(guang)调控大(da)肠杆菌中的蛋白表达,并且可以通过光(guang)释(shi)放巴氯芬药物分子(zi)有效(xiao)调控GABAB受(shou)体(ti)功(gong)能,进(jin)(jin)而对小(xiao)鼠脑片中的(de)神经细胞膜(mo)电位进(jin)(jin)行高时空分辨率的(de)光调控(kong)。最后使(shi)用线粒体(ti)定(ding)位罗丹明衍生(sheng)物染料(liao)作为具有(you)定(ding)位效应(ying)的(de)光催化剂,借助于过氧(yang)化氢在(zai)细胞内的(de)有(you)限自由(you)扩散,实现了(le)亚细胞定(ding)位的(de)选择性小(xiao)分子可见(jian)光去笼(long)。该研究展示(shi)了(le)光催化氧(yang)化还原反应(ying)在(zai)细胞及(ji)生(sheng)命(ming)体(ti)系研究中的(de)广阔应(ying)用前景,并提出了(le)有(you)机荧光染料(liao)具有(you)示(shi)踪与(yu)调控(kong)生(sheng)命(ming)过程的(de)双重功(gong)能,对相关领域的(de)研究具有(you)重要启(qi)示(shi)。

      上述(shu)研(yan)究工作(zuo)得到国(guo)(guo)家(jia)重大科(ke)学研(yan)究计划、国(guo)(guo)家(jia)自然科(ke)学基(ji)金(jin)委(wei)、中科(ke)院战略(lve)性(xing)先导科(ke)技专项(xiang)(B类)、上海市重点项目、生命(ming)有机(ji)化学国家重(zhong)点(dian)实验室及中(zhong)科院的资助。
